[Animal Modeling - Pharmacological Evaluation] - Chronic Renal Failure Rat Model Induced by Xiaozhiling

  1. Modeling material animals: Male Wistar rats weighing around 200g; Drug: Refined Xiaozhiling Injection.

  2. Modeling method: The modeling animals are first anesthetized with ketamine and diazepam, and if necessary, supplemented with ether inhalation. Rats were fixed in a supine position, and a 1.2ml dilution of refined Xiaozhiling injection was prepared by adding an equal amount of physiological saline to 0.6ml of Xiaozhiling injection and injected into both kidneys. The injection method is to insert the needle from the lower pole (4th needle) to the upper pole of the middle pole, as much as possible towards the cortex, with the needle tip facing the cortex. Slowly inject (at least 1 minute each time), and press the needle eye with an iodine swab for more than 3 minutes after exiting the needle.

  3. The principle of mold making: Xiaozhiling is mainly composed of effective ingredients such as traditional Chinese medicine Wubeizi and alum. The main component of Wubeizi, tannic acid, has strong tissue astringency, which can cause protein coagulation and vascular constriction. The main component of alum, potassium aluminum sulfate, has a strong inflammatory effect on the local area and causes tissue fibrosis. Injecting an appropriate amount of Xiaozhiling into the kidneys can cause damage to most renal units, leading to the pathological process of CRF.

  4. Changes after modeling: Within 3 days after modeling, animals should drink and eat less, resulting in weight loss. After one week, the weight will increase, and there will be symptoms such as pale tail and ears, fluffy hair, uneven dryness, and hair loss. After 16 weeks, the weight of the modeling group was significantly lighter than that of the normal control group.

  5. Biochemical and pathological changes after modeling: Within one week after modeling, there may be varying degrees of hematuria, with proteinuria mostly above+++, which continues to fluctuate between+and++++after one week. Scr and BUN show a progressive upward trend.

  The pathological process shows compensatory hypertrophy of glomeruli, proliferation and dilation of renal tubules, and late stage; Metabolic disorders, severe damage to glomerular and tubular functions, and high consistency with pathological changes in human CRF with renal atrophy were observed under both light and electron microscopy.
