[Animal modeling - efficacy evaluation] - establishment of diabetes animal model after total pancreatectomy

  【 Operation steps 】 Adult hybrid dogs weighing 10-15kg are injected with 30mg/kg body weight of 3% pentobarbital sodium via intramuscular injection and tracheal intubation. One side of the femoral artery and vein were respectively intubated to measure arterial pressure and collect blood samples. After anesthesia, intravenous infusion of Ringer's solution was administered. After laparotomy, the pancreas was found, and the main supply vessels of the canine pancreas were the splenic artery and the superior and inferior pancreaticoduodenal arteries. The latter forms an anastomotic arch at the upper edge of the pancreas and emits many small blood vessels to supply the duodenum, pancreatic head, and body. The duodenal recurrent artery branches off from the gastroduodenal artery and supplies the proximal duodenum. During the operation, the pancreaticoduodenal anastomosis arch is first dissected. At the lower edge of the anastomosis arch, all small blood vessels that originate from the pancreas are ligated and cut one by one with ophthalmic fine wires, gradually freeing the pancreas. Near the gastroduodenal artery, the anastomotic arch may pass through the pancreatic parenchyma, so the pancreas above it must be removed together. Then, the pancreatic duct is separated from the pancreatic body and its distal end is tightly sutured after cutting to prevent intestinal fistula. Finally, cut off the main supply blood vessels to minimize their ischemia time as much as possible. Then place the dog in a cage and feed it with pancreatic enzyme tablets to supplement insufficient exocrine secretion.

  [Result analysis] The blood glucose rose to above 11.1 mmol/L within 3 days after total pancreatectomy, and there were symptoms of polydipsia, polyuria, emaciation, and even diabetes ketoacidosis.

  【 Precautions 】 Wrap the animal with a towel to keep it warm after surgery, and place the animal outside the cage until fully anesthetized and then put it in a dry cage. Administer physiological saline to animals within 5 days after surgery, and remove stitches on the 7th day after surgery.
