[Animal Modeling - Pharmacological Evaluation] - Pathological Tongue Coating Model in Small Pigs

  The changes in tongue coating and tongue texture are one of the main bases for clinical differentiation in traditional Chinese medicine. If similar changes to human tongue coating and tongue texture can be replicated in animal tongues, it will contribute to the development of animal models for traditional Chinese medicine "syndrome". Due to the significant differences in tongue structure between rats, guinea pigs, cats, rabbits, and dogs and human tongue, the tongue structure of macaques, although similar to human tongue, is expensive and limited in application. The tongue structure and diet of miniature pigs are similar to those of humans, and they have been widely used in experimental research in various disciplines of medicine. They are more suitable as experimental animals for traditional Chinese medicine pathological tongue images and "syndrome" models.

  1. Experimental method: Xishuangbanna miniature pig, male, weighing 21-22.5kg. A 1:1 mixture of 50% Baijiu and saturated edible alkali solution was used to make the model by gavage every other day. Gastric lavage is performed around 9am in the morning, followed by feeding with regular full price feed in the afternoon. Fasting for 24 hours before gavage is repeated for about 50 days.

Experimental results: This study successfully replicated a relatively long-lasting thin white greasy tongue coating based on the pathogenesis of TCM's external cold and dampness and internal damage to the spleen and stomach, which can easily lead to greasy coating. From the the fourth day of the experiment, miniature pigs began to appear and maintain a relatively durable thin white and greasy tongue coating. The tongue coating presents a thin white and slightly dry appearance, with a pale red and slightly dark tongue texture. Through scanning electron microscopy observation, it was found that the ultrastructure of the tongue surface of miniature pigs is basically similar to that of humans. Transmission electron microscopy examination showed that the cell structure and characteristics of each layer of the tongue epithelium in miniature pigs were basically the same as those of human tongue epithelial cells. From the perspective of anatomy and histology of tongue structure, miniature pigs fully meet the requirements for making animal models of tongue coating in traditional Chinese medicine pathology.
