[Animal Modeling - Drug Efficacy Evaluation] - Traditional Chinese Medicine Exercise Fatigue Animal Model

  (1) Establishing a spleen qi deficiency animal model using fatigue factors and laxative drugs such as rhubarb and saltpeter

  The replication method used male Wister rats weighing 180-200g. Mix rhubarb, saltpeter, and standard feed evenly in a ratio of 0.85:0.15:9.0 to make medicinal feed. The experimental animals were fed medicated feed daily and vibrated on a fatigue device oscillator (243 oscillations/min, amplitude of 36mm) for 4 hours from 8:00 to 12:00 in the morning, with a modeling period of 21 days. Observe the appearance, feces, body weight, food intake, urination, and defecation of rats, and take blood to measure the levels of red blood cell C3b receptors, IgG, and IgM; Take the spleen to measure T cell subsets, TL22 activity, NK activity, and lymphocyte transformation rate.

  (2) Establishing an animal model of spleen qi deficiency using overwork and senna leaves

  The replication method used Kunming white mice weighing 20-30g, and a model was established by combining senna with swimming exercise. Perform swimming training once a day for 2-5 minutes, with the body sinking as the degree, and apply 0.5ml/piece of 100% senna decoction. On the fourth day of modeling, the experimental animals showed squinting, laziness, clumping, soft stools in stripes, loose and dull fur, and obvious timidity. On the 8th day of modeling, the patient was euthanized and blood was collected to measure the levels of red blood cell C3b receptors, IgG, and IgM; Take the spleen to measure T cell subsets, TL-2 activity, NK activity, and lymphocyte transformation rate.

  (3) The characteristic of the traditional Chinese medicine animal model of exercise-induced fatigue is to apply a load of exercise on top of diarrhea, causing a decline, imbalance, and insufficient blood and essence in the animal's internal organs. In the above two models, a significant decrease in the appearance and immunological indicators of the animals indicates that they have developed spleen qi deficiency syndrome.
