【 Modeling Method 】 Healthy female or male rats aged 3-12 months were selected, and dexamethasone (1.0-2.5mg/kg) was injected into the gluteal or hind leg muscles twice a week. After 6-8 weeks, a glucocorticoid induced osteoporosis model could be replicated. The control group received intramuscular injection of physiological saline injection, with the same injection method as the experimental group.
After 8 weeks of modeling, the bone structure showed sparse and small trabeculae, thinning thickness, and disordered arrangement. This model can be accelerated by a low calcium diet, and the application of vitamin D3 can prevent its formation.
With the widespread use of glucocorticoids in clinical practice, the side effects of osteoporosis caused by glucocorticoids have gradually become a focus of attention. This model is mainly used to simulate secondary osteoporosis caused by human glucocorticoids and has positive significance for studying secondary osteoporosis caused by human glucocorticoids. Corticosteroids directly inhibit the function of osteoblasts, reduce the transformation of pre osteoblasts into functional osteoblasts, decrease the synthesis of collagen in pre osteoblasts, and reduce the formation of bone matrix. The pathogenesis and progression of clinical glucocorticoid induced osteoporosis and primary osteoporosis are not consistent, so the glucocorticoid induced osteoporosis model is not suitable for studying the inhibitory effect of drugs on bone resorption, such as parathyroid hormone, prostaglandin, calcitonin, and estrogen.
Corticosteroid induced osteoporosis models are commonly used in rats, with simple methods and high success rates, and are more widely used than other animals.
