【Animal Modeling - Pharmacological Evaluation】 - Rabbit Masugi Nephritis Model

  1. Modeling material animals: rabbits, weighing 2-2.5kg, sheep; Drug: Complete Freund's adjuvant.

  2. The modeling method involves preparing sheep anti rabbit anti kidney serum in advance. Take rabbits, regardless of gender, and euthanize them. Remove the kidneys, insert a catheter, and rinse them repeatedly with physiological saline. Take 5g of renal cortex and grind it into a homogenate, mix it with complete Freund's adjuvant to make 10ml, and add 20ml of physiological saline to make a suspension. Inject subcutaneously or intramuscularly into sheep in 5 locations, with 1ml per location, once every 2 weeks, for a total of 4 times. Two weeks after the last injection, sheep blood was collected to isolate serum, and equal amounts of rabbit red blood cells were washed three times with physiological saline and mixed well. Leave it overnight at 4 ℃ to allow it to fully adsorb, centrifuge and discard blood cells that have already adsorbed foreign antibodies. Extract the supernatant and inactivate it in a 56 ℃ water bath for 0.5 hours for future use, which is anti renal serum.

  Inject the above-mentioned anti renal serum intravenously into rabbits, 1.5-2ml each time, every half hour, about 3-8 times, until proteinuria occurs.

  3. The principle of modeling is to induce an animal model of glomerulonephritis using anti renal serum method.

  4. Changes after modeling: If proteinuria appears after modeling, it indicates successful modeling.
