1. Animal model material: Dog, weighing over 10kg; Medications: pentobarbital, physiological saline, lidocaine; Equipment: Animal artificial respiration machine.
2. Method of modeling: After anesthesia with pentobarbital, the animal is fixed in an upward position, and the trachea is cut open to connect the animal's ventilator for artificial respiration. The chest is opened in the fourth intercostal space on the left, exposing the heart, and cutting the open capsule. A silk thread is used to ligate the left anterior descending branch (LAD) at the upper one-third (1-2cm from the starting point of the LAD), 1/2, or 1/3 (above the apical branch) of the left coronary artery, resulting in myocardial infarction. This is a one-stage ligation method. To reduce the occurrence of arrhythmia and mortality in animals, the two-stage ligation method is commonly used, which involves threading two silk threads at a ligation point, placing the 5th needle parallel to the coronary artery in the ligation node, ligating the first silk thread (the first node), ligating the needle together with the anterior descending branch of the coronary artery, and extracting the needle; After 30 minutes, ligate the second silk thread (second ligation); At this point, the anterior descending branch of the left coronary artery is completely ligated. If an epicardial electrode is not used, the pericardium can be sutured, the chest wall can be sutured, and natural respiration can be restored. An electrocardiogram oscilloscope can be connected to observe the changes in electrocardiogram. Physiological saline can be dripped from the femoral vein and external jugular vein according to changes in blood loss.
3. The principle of modeling is to replicate a myocardial infarction model by ligating the coronary artery.
4. Pathological changes after modeling: After modeling, the animal's electrocardiogram showed ST segment elevation and pathological Q-wave, indicating the occurrence of myocardial infarction.
5. Precautions: Surgical instruments should be strictly disinfected to prevent surgical infections. Surgical trauma should be minimized and sterile operations should be strictly carried out.
