[Animal modeling] - Escherichia coli endotoxin induced rat uveitis model

  1. Model making method: Healthy purebred Wister rats, weighing around 200mg, with endotoxins (ET): the effective component of ET, lipopolysaccharide (LPS), extracted from Escherichia coli with a purity of 99%. Before use, they were dissolved in physiological saline to form a 2mg/ml solution. Injection of LPS 600 into the left foot pad of model group rats μ G/kg.

  2. Observation indicators and analysis

  (1) Aqueous humor measurement: Inject LPS for 24 hours, extract the aqueous humor from 3mm behind the rat corneal limbus, and immediately send it for examination to detect the number of white blood cells and protein content in the aqueous humor.

  (2) Eye manifestations: Before the experiment, the eyes of rats were examined using a slit lamp and ophthalmoscope. After LPS injection, the eyes were examined every 6 hours, and the results of the 24-hour examination after the experiment were used as experimental eye signs. The examination includes the conjunctiva, cornea, aqueous humor, iris, lens, vitreous body, and retina of both eyes.

  (3) Pathological examination: There are a large number of inflammatory cells such as neutrophils, lymphocytes, and monocytes in the iris tissue, and there is cellulose like exudation in the anterior chamber.
