【Animal modeling】 - animal model of liver qi stagnation syndrome caused by bandage restraint

  (1) Reproduction method: Adult rats were placed in a breeding cage, and their limbs were bound with a thin bandage to make it difficult to walk. The state of the rats was observed. After one week, changes in liver tissue, plasma malondialdehyde (MDA), liver tissue, and red blood cell superoxide dismutase (SOD) were detected, and the ultrastructure of liver cells was observed under electron microscopy.

  (2) The characteristics of the model showed typical appearance changes 3 to 4 days after animal modeling; Plasma and liver tissue MDA significantly increased; Red blood cells and liver tissue SOD significantly decreased; Under electron microscopy, the mitochondria of liver cells show significant swelling, reduced matrix, partial dissolution of mitochondrial membranes, unclear internal and external membrane structures, blurred nuclear membranes, reduced chromatin, cell membrane fusion, and severe detachment of intracellular components.

  (3) Comparative medicine liver qi stagnation syndrome refers to a syndrome characterized by liver dysfunction and qi stagnation, also known as liver qi stagnation syndrome. Its main clinical features include emotional depression, chest and hypochondriac or abdominal distension, pain, or menstrual disorders in women. With the acceleration of life rhythm and the increase of life pressure, people are increasingly subjected to various external pressures. Emotional disorders and liver qi imbalance have become common causes in clinical practice. Emotional modeling causes psychological stress reactions in animals, which is related to the etiology of liver depression in traditional Chinese medicine. However, there are still three issues worth further exploration. One is the relationship between "anger injuring the liver" and liver depression. Anger injuring the liver can lead to liver depression, but it can also lead to hyperactivity of liver yang; Which type of syndrome appears depends on the amount of stimulation or animal strain? Secondly, not all liver depression in traditional Chinese medicine is caused by anger. All emotional disorders and the absence of depression can lead to liver depression syndrome. For example, excessive social pressure or frustration, and sadness without a place to express can also lead to liver depression syndrome, which is commonly seen in clinical practice. Therefore, there are some differences between the liver depression model caused by anger damaging the liver and the liver depression model that appears in clinical practice; Thirdly, the performance of liver depression models varies, and there is currently no unified evaluation standard. The basic pathological change of liver depression syndrome is liver loss and laxity. Based on the analysis of currently available data, liver depression syndrome involves multiple functional changes in the nervous system, digestive system, endocrine system, blood circulation system, and other systems. When studying these pathological changes, emotional stimulation modeling method is mostly used. Compared with the blank control group, there are significant differences in the changes of various indicators or the drug has a good anti evidence effect, indicating that these pathological changes are universal in liver depression syndrome, However, there is a lack of specificity, and many other syndrome types also have pathological changes in these aspects, which require further in-depth research.
