【Animal Modeling】 - Reflux Gastritis Model

  1. Animal modeling materials: Male rats, weighing 200-250g; Medications: pentobarbital, physiological saline, iodine alcohol.

  2. Modeling method

  (1) Model rats were anesthetized by intraperitoneal injection of pentobarbital at a dose of 30mg/kg, followed by skin disinfection with iodine. After laparotomy, the stomach and upper jejunum were exposed, and the anterior stomach was fixed together with a jejunum 5-6cm away from the pylorus. A 7-8mm incision was left in the middle. First, a 6-7mm incision was made in the anterior stomach, and the gastric cavity was carefully washed clean. Attention was paid to not flushing the gastric contents into the abdominal cavity. Then, a small incision of the same size was made in the small intestine, First, intermittently suture the inner side of the stomach and small intestine incision (using No.0 silk thread), then suture the outer side, seal the incision, and complete the anastomosis. Rinse the abdominal cavity twice with sterile physiological saline, and dry it with clean gauze. Then, suture the abdominal wall muscle layer and skin layer by layer, and close the abdominal cavity. Postoperative fasting for 1 day and modeling for 3 months.

  (2) Male SD rats were anesthetized with pentobarbital (30mg/kg, intraperitoneal injection). Cut off the abdominal hair, disinfect the skin, open the upper abdomen, expose the stomach and upper jejunum, and fix the anterior stomach with the jejunum 5-6cm away from the pylorus. Leave 7-8mm in the middle for all openings. Firstly, cut a 6-7mm incision in the anterior stomach, carefully rinse the gastric cavity with physiological saline, and then cut a small incision of the same size into the small intestine. First, sew the inner side of the stomach and small intestine incision, then sew the outer side, seal the incision, rinse the abdominal cavity twice with physiological saline, dry with disinfectant gauze, and inject 2-3 drops of gentamicin dilution. Suture the muscle layer and skin layer by layer, close the abdominal cavity, and then add 2-3 drops of gentamicin diluent, fasting for 24 hours.

  3. Modeling principle: Surgery causes reflux gastritis in animals.

  4. After 3 months of modeling, pathological sections showed significant infiltration of inflammatory cells, glandular hyperplasia, and intestinal metaplasia; To the naked eye, the gastric mucosa is congested, congested, and arranged in a cobblestone shape.

  5. The main pathological changes after modeling occur in the mucosa around the gastrointestinal anastomosis, including infiltration of inflammatory cells, atrophy of gastric glands, intestinal metaplasia of gastric mucosa, and width of gastric pits. Inflammation is characterized by the highest number of lymphocytes, most of which occur in the lamina propria of the mucosal layer. Some specimens have infiltrated into the mucosal muscle layer and submucosa.
