(1) Reproduction method: Adult rats were gavaged with 4 ℃ vinegar daily, with a volume of 10ml/kg body weight for 10 consecutive days. During this period, observe changes in animal appearance and behavior, weight, food intake, and water intake. On the day after the completion of modeling, gastric emptying and gastrointestinal propulsion tests were conducted. After blood collection, all animals were sacrificed and all gastrointestinal tracts were taken. The residual rate of gastric substances was calculated by weighing the total and net weights of the stomach; Detect the levels of motilin (MTL), gastrin (GAS), superoxide dismutase (SOD), and glutaraldehyde (MDA) in serum and gastric mucosal tissue. The remaining animals continued to be fed, and their body weight and food intake were regularly measured. On the 7th day after the cessation of modeling, gastric emptying and propulsion rates were measured, and blood and gastric mucosa were taken for the above examinations.
(2) The characteristics of the model include reduced animal activity, manifested as fatigue and lack of movement, slow response, pale lips, weak resistance during capture, and gradually becoming thin and weak, with loose stools and weight loss. The amount of food and water consumed is significantly reduced, but it still does not recover after stopping the modeling. At the end of modeling, the residual rate of substances in the stomach increased, indicating a slowing down of gastric emptying. It continued to exist and worsen one week after stopping modeling, and its impact on the small intestine propulsion rate of animals was also significant. After modeling, the SOD activity in the stomach and blood of the animals decreased, the MTL content decreased, the MDA content in the gastric mucosa increased, and it did not recover after 1 week of stopping modeling. There was no significant change in the GAS content in the stomach and blood.
(3) Comparative medicine spleen stomach deficiency cold syndrome refers to the deficiency of spleen qi in the body, or the prolonged spleen deficiency, which affects the spleen yang and leads to the deficiency of the spleen yang; Or eating too much to produce cold, damaging the spleen yang, causing spleen yang deficiency to not warm the stomach and intestines, and cold qi originating from within. The main symptoms are stomach pain or dull abdominal pain, like to warm and press, very painful on empty stomach, less pain after eating, suffused with water, poor appetite, low spirit, fatigue, cold hands and feet, loose stool, pale tongue coating, weak or slow pulse. The spleen and stomach deficiency cold model prepared by vinegar method seems to be closer to the clinical manifestations of spleen and stomach deficiency cold in traditional Chinese medicine, and the changes in gastrointestinal hormones and other related indicators are relatively stable, based on the general changes and related test results of the animals during and after the model is stopped. This model first combines the theory of traditional Chinese medicine on the formation of deficiency syndrome, which is more sufficient in terms of modeling time. The theory that overeating sour taste can cause spleen deficiency has been discussed as early as in the Yellow Emperor's Internal Classic, and in addition, a large amount of cold vinegar is given for a long time, which together constitute the conditions for causing spleen and stomach deficiency cold. Therefore, the prepared animal model of spleen and stomach deficiency cold is more in line with traditional Chinese medicine theory and clinical practice.
