[Animal modeling] Urine metabolomics of type 2 diabetes rats with qi yin deficiency syndrome

  Objective To explore the intrinsic biomaterial basis of Chinese medicine syndromes of type 2 diabetes in adult SD rats from the aspects of metabolic differential markers and metabolic pathways through urine non targeted metabonomics.

  Methods SD rats were randomly divided into normal group and type 2 diabetes mellitus (T2DM) group with deficiency of both qi and yin. Non target metabolomics analysis of urine was performed using Liquid Chromatography Mass Spectroscopy (LC-MS) technology. Principal Component Analysis (PCA) and Partial Least Squares Discriminant Analysis (PLS-DA), KEGG database, HMDB database The LIPIDMaps database and liquid chromatography tandem mass spectrometry (LC-MS/MS) were used to identify potential biomarkers for pathway enrichment analysis.

  The results showed that the metabolic changes related to diabetes involved 9 carbohydrate metabolism, 12 lipid metabolism, 9 amino acid metabolism and 2 energy metabolism.

  Conclusion There are carbohydrate metabolism, lipid metabolism, amino acid metabolism and bioenergy metabolism disorders in the biological model of qi yin deficiency of diabetes. Metabonomics research methods are helpful to reveal the microscopic material basis of TCM syndromes.
