[Animal modeling] - Xiaoyusan new formula for repairing cartilage damage and MMP-13 expression in knee Osteoarthritis model rabbits

  Objective To study the effect and mechanism of Xiaoyusan new formula on articular cartilage of knee Osteoarthritis rabbits.

  Method: 42 6-month-old ordinary grade New Zealand white rabbits were randomly divided into a normal group, a model group, a Xiaoyu San group, and a new Xiaoyu San formula group, with 10 rabbits in each group (the other 2 rabbits were used for model validation). Except for the normal group, all other groups of rabbits underwent KOA modeling of the right knee joint using the improved Hulth method. After 5 weeks of modeling, the animals in the Xiaoyu Powder group and the Xiaoyu Powder New Formula group were treated with corresponding patches for knee osteoarthritis, once a day, for 10 hours each time. After 2 weeks of continuous medication treatment, four groups of rabbit knee joint cartilage were taken for gross observation and evaluated for cartilage damage using the Outerbridge grading method. HE staining was used to observe the pathological changes of knee joint cartilage, calcified layer, and subchondral bone in each group under an optical microscope, and the degree of cartilage degeneration was evaluated using the Mankin's method, The expression of Matrix metalloproteinase 13 (MMP-13) in the articular cartilage of rabbits' knees in each group was observed by immunohistochemical method under the light microscope.

  After gross observation of the articular cartilage, the Outerbridge rating comparison showed that the number of high-grade animals in the Xiaoyu San group decreased compared to the model group (P<0.05), and the number of high-grade animals in the Xiaoyu San new formula group also decreased compared to the Xiaoyu San group (P<0.05). HE staining revealed that the Mankin's scores of the articular cartilage in the four groups of rabbits were ranked from high to low in the model group (10.82 ± 1.76), Xiaoyu powder group (6.19 ± 1.23), Xiaoyu powder new formula group (2.64 ± 1.18), and control group (0.28 ± 0.17), with statistically significant differences between the groups (P<0.05). Immunohistochemical detection showed that the positive rate of MMP-13 expression in the articular cartilage tissue of rabbits in each group, from high to low, was 67.90 ± 13.94% in the model group, 37.10 ± 19.16% in the Xiaoyu Powder group, 13.60 ± 3.10% in the Xiaoyu Powder new formula group, and 3.20 ± 2.39% in the control group. The differences between the groups were statistically significant (P<0.05).

  Conclusion Xiaoyusanxin recipe can repair the articular cartilage injury of KOA rabbits and reduce the expression level of MMP-13 in cartilage tissue, which may be one of its mechanisms in treating Osteoarthritis.
